Introduction
Acid-Washed glass beads are specifically designed for grinding and lysing fungi, with a diameter of 0.4-0.6mm (30-40 mesh). When glass beads are added to the solution with high-speed vortexing or oscillation , the fungal cell wall can be efficiently removed. In nucleic acid extraction, many fungi have unique cell wall structures, and lysozyme may not be effective. In this case, acid washed glass beads can be used as grinding media to lyse bacterial through high-speed vortexing or oscillation. This method is also called bead grinding. Acid-washed refers to soaking glass beads in high concentrations of hydrochloric acid and nitric acid to remove residual metals and metal oxides, in order to prevent these substances from affecting the quality of nucleic acid.
Usage
In a 2ml centrifuge tube, add 300-500μl homogenate and 200-300mg acid-washed glass beads (0.4-0.6mm), vortex at maximum speed for 5-10 minutes.
